Negative Feedback by NUR77/ Nr4a1 Restrains B Cell Clonal Dominance During Early T-Dependent Immune Responses

چکیده

B cell clones compete for entry into and dominance within germinal centers (GC), where the highest affinity BCRs are selected. However, diverse low cells can enter reside in GCs extended periods. To reconcile these observations, we hypothesized that a negative feedback loop may operate to preferentially restrain high from monopolizing early GC niche. Here report role nuclear receptor NUR77/Nr4a1 this process. We previously showed NUR77 expression scales with antigen stimulation restrains expansion when T help is limiting. show that, although dispensable regulating size elicited largely clonal manner, it serves curb immunodominance under conditions populations must constrained Moreover, independent of precursor frequency reflects, at least part, cell-intrinsic NUR77. propose important preserve diversity order limit holes post-immune repertoire optimize selection.

B Cell–dependent T Cell Responses

Contact sensitivity (CS) is a classic example of in vivo T cell immunity in which skin sensitization with reactive hapten leads to immunized T cells, which are then recruited locally to mediate antigen-specific inflammation after subsequent skin challenge. We have previously shown that T cell recruitment in CS is triggered by local activation of complement, which generates C5a that triggers C5a...

Exclusion of natural autoantibody-producing B cells from IgG memory B cell compartment during T cell-dependent immune responses.

In healthy individuals, a substantial proportion of circulating Abs exhibit polyreactivity and self-reactivity. These Abs are referred to as natural autoantibodies (NAAs). As part of the innate immunity, NAAs play an important role in eliminating pathogens. However, inherent to their poly/autoreactivity is the potential for NAAs to differentiate to high-affinity autoantibodies during an immune ...

IRF2BP2 transcriptional repressor restrains naive CD4 T cell activation and clonal expansion induced by TCR triggering.

CD4 T cell activation and differentiation mechanisms constitute a complex and intricate signaling network involving several regulatory proteins. IRF2BP2 is a transcriptional repressor that is involved in gene-expression regulation in very diverse biologic contexts. Information regarding the IRF2BP2 regulatory function in CD4 T lymphocytes is very limited and suggests a role for this protein in ...

B cells enhance early innate immune responses during bacterial sepsis

Microbes activate pattern recognition receptors to initiate adaptive immunity. T cells affect early innate inflammatory responses to viral infection, but both activation and suppression have been demonstrated. We identify a novel role for B cells in the early innate immune response during bacterial sepsis. We demonstrate that Rag1(-/-) mice display deficient early inflammatory responses and red...
